摘要:
九层塔(Ocimum basilicum L )为一年生唇形科药食两用植物。为研究野生九层塔植株内生细菌的多样性及生物学功能,采用LB、VM、DN和CCM四种培养基从植物不同部位中分离纯化内生细菌,利用IS-PCR插入序列指纹图谱技术进行聚类分析,通过16S rRNA基因测序分析对内生细菌进行分子鉴定及系统发育分析,进一步采用传统方法鉴定各类群代表菌株的生物学功能,并选取功能性较强的菌株进行水稻种子萌发及盆栽促生试验。结果显示,从九层塔中分离获得48株内生细菌,聚为13个类群,表现出九层塔内生菌的多样性;生物学功能测定显示所获得内生菌功能丰富:9株具有固氮功能,8株具溶磷功能,10株具有解钾能力,12株具有分泌生长素的能力,7株具有分泌铁载体功能;筛选出2个促生功能优良的菌株,分别为H1041和H4039,其发酵液能显着地促进水稻种子萌发和水稻生长。本研究表明从野生植物九层塔中分离所得的内生菌具有种群多样性和功能多样性,这些兼具不同功能的菌株具有农业应用开发的价值和意义。(图6 表3 参62)
Abstract:
Ocimum basilicum L is an annual medicinal and edible plant of Labiatae. In order to investigate biodiversity and physiological function of endophytic bacteria, the plant Ocimum basilicum L. was chosen as experiment material. Endophytic bacteria were isolated and purified from the plant by LB, VM, DN and CCM mediums. Genetic diversity of the isolates were studied by insertion sequence-based PCR (IS-PCR) finger-printing. The 16S rRNA gene phylogenetic analysis and physiological and biochemical tests were used to identify the representative strains of all clusters. The biological functions of representative strains of various groups were further identified by traditional methods, and the strains with strong functions were selected for further rice seed germination and pot culture. As a result, 48 endophytic bacteria were isolated and purified from the Ocimum basilicum L, which were clustered into 13 groups with IS-PCR fingerprinting method, showing the diversity of endophytic bacteria in the Ocimum basilicum L. Biological function tests showed that the endophytic bacteria had abundant functions ,in which 9 strains had the function of nitrogen fixation, 8 solubility function, 10 potassium solubility, 12 auxin secretion, and 7 sideophore secretion. Two strains with excellent functions, H1041 and H4039, were screened out. Their fermentation could both significantly promote the germination of rice seeds and the growth of rice. This experiment showed that the endophytic strains isolated from Ocimum basilicum L. have abundant t diversity both in types and functions.
